Unpacking extracellular vesicles: RNA cargo loading and function

چکیده
Extracellular vesicles (EVs) are a heterogeneous group of membrane-enclosed structures produced by prokaryotic and eukaryotic cells. EVs carry range biological cargoes, including RNA, protein, lipids, which may have both metabolic significance signalling potential. EV release has been suggested to play critical role in maintaining intracellular homeostasis eliminating unnecessary material from producing cells, as delivery system enable cellular communication between neighbouring distant cells without physical contact. In this review, we give an overview what is known about the relative enrichment different types RNA that associated with most recent research efforts. We then examine selective non-selective incorporation these biotypes into EVs, molecular systems sorting elucidated so far, process EV-producing Finally, also discuss model providing evidence for EV-mediated recipient implications relevance physiological pathological scenarios.

Low active loading of cargo into engineered extracellular vesicles results in inefficient miRNA mimic delivery
Extracellular vesicles (EVs) hold great potential as novel systems for nucleic acid delivery due to their natural composition. Our goal was to load EVs with microRNA that are synthesized by the cells that produce the EVs. HEK293T cells were engineered to produce EVs expressing a lysosomal associated membrane, Lamp2a fusion protein. The gene encoding pre-miR-199a was inserted into an artificial ...
متن کاملEXTRACELLULAR VESICLES AND THEIR THERAPEUTIC APPLICATIONS: A REVIEW
Background & Aims: Extracellular vesicles are double phospholipid layer vesicles releasing from several cells. These particles contain bio-molecules including mRNA, miRNA, DNA, proteins, and lipids which play a pivotal role in the cell to cell communication. Once delivered, they participate to change target cell function, fate, morphology, differentiation, and growth. The present study is a re...
متن کاملThe origin, function, and diagnostic potential of RNA within extracellular vesicles present in human biological fluids
We have previously demonstrated that tumor cells release membranous structures into their extracellular environment, which are termed exosomes, microvesicles or extracellular vesicles depending on specific characteristics, including size, composition and biogenesis pathway. These cell-derived vesicles can exhibit an array of proteins, lipids and nucleic acids derived from the originating tumor....
متن کاملIsolation and characterization of Faecalibacterium prausnitzii extracellular vesicles
Introduction: Extracellular vesicles (EVs) contain active biological compounds which play important roles in biological processes. The secretion of EVs is a common phenomenon which occurs in archaea, bacteria and mammalian cells. The secretion of bacterial EVs has been discovered in various species of both Gram-negative and Gram-positive bacteria. Faecalibacterium prausnitzii is one of the comm...
متن کاملLysozyme association with circulating RNA, extracellular vesicles, and chronic stress
BACKGROUND Stress has demonstrated effects on inflammation though underlying cell-cell communication mechanisms remain unclear. We hypothesize that circulating RNAs and extracellular vesicles (EVs) in patients with chronic stress contain signals with functional roles in cell repair. METHODS Blood transcriptome from patients with Irritable Bowel Syndrome versus controls were compared to identi...
